LOST: Season 3 Finale - was a gamechanger (but there are a few other really good ones like the one where we find out Ben's story).

Seinfeld: 1) Not that there's anything wrong with that
2) Poppy's first episode

I'm sure there will be more shows that come to mind later.

I'm sure I could come up with hundreds , but the Lost pilot episode was pretty amazing. I've never felt so on the edge of my seat than I was for the first ten or fifteen minutes of that.
